What harvest titer should I assume for a CHO mAb feasibility?
Doing an early feasibility for a monoclonal antibody in CHO fed-batch. Before we have any cell-line data, what titer do you plug in for a first-pass plant sizing — a conservative 2–3 g/L, or the 5+ g/L modern platforms hit? How much margin do you leave?

For a first-pass feasibility before you have any clone data, 2.5–3 g/L is a sensible, defensible planning number for a modern CHO fed-batch platform — realistic but not over-promising. Hold 5+ g/L for when you actually have platform/clone data behind it. Practical tip: size at the conservative titer, then run the sensitivity — roughly, halving titer doubles the required working volume, so you see immediately how exposed the design is. We also keep ~20–30% margin on top for failed batches and ramp-up.
